Description
English: Immunofluorescence staining of the primo-vessel in a lung cancer xenograft.

Primo-vessel cross sections were treated with anti-LYVE1 polyclonal antibody (green) or iso-type control polyclonal antibody (C, D) and observed with confocal microscopy. Lymphatic vessel cross sections were stained with the same antibodies and observed as positive controls (A, B). Fluorescence images of DAPI (blue) exhibiting nuclei and DIC images were also taken. Lymphatic endothelial cells with strong positive staining clearly located at the inner boundary of lymphatic vessel, whereas no LYVE-1 positive cells were seen in the primo-vessel. A part of the same sample which was treated with only DAPI without sectioning and mounted longitudinally revealed aligned rod-shape nuclei surrounding lumens assumed to be endothelial cells of primo-vessel (E).

Deutsch: Immunofluoreszenzfärbung des Primogefäßes bei Lungenkrebs (Xenotransplantate) am Mausmodell Die Querschnitte von Primogefäßen wurden mit polyklonalen anti-LYVE1 Antikörpern (grün) oder isotypen polyklonalen Kontrollantikörpern (C, D) kontrastiert und mittels Konfokalmikroskopie untersucht. Die Querschnitte von Lymphgefäßen wurden gleichermaßen behandelt und als positive Kontrollen verwendet (A, B). Fluoreszenzbildgebung mit DAPI, einem Farbstoff, der besonders in der DNA bindet, die reich an Adenein und Thymin sind und digitale Bildkorrelation wurden eingesetzt. Die lymphatischen endothelialen Zellen färbten sich stark positiv an und waren klar an der inneren Begrenzung des Lymphgefäßes lokalisiert. Hingegen fanden sich keine LYVE-1 positiven Zellen im Primogefäß. Ein Teil der gleichen Probe, die nur mit DAPI behandelt worden war ohne sie aufzuschneiden und in Längsrichtung gelagert wurde, wies ausgerichtete stabförmige Kerne um Hohlräume herum auf, von denen anzunehmen ist, daß es sich um endotheliale Zellen von Primogefäßen handelt (E).
