Поляризаційна мікроскопія відрізняється від класичної світлової мікроскопії використанням двох поляризаційних фільтрів – поляризатора і аналізатора, що розташовуються між джерелом світла і мікропрепаратом (поляризатор) та між об’єктом і об’єктивом (аналізатор). Обидва поляризаційні фільтри забезпечують поляризацію світла у певній площині, що називається площиною поляризації. Коли площини поляризації поляризатора і аналізатора співпадають, поляризоване світло, що пройшло через поляризатор, проходить через мікропрепарат і безперешкодно проходить через аналізатор (А). Коли площини поляризації поляризатора і аналізатора взаємно перпендикулярні, світло, що пройшло через поляризатор (поляризоване у площині поляризатора), відсіюється аналізатором, обумовлюючи темний фон. Однак якщо між поляризатором і аналізатором є об’єкти із анізотропними властивостями, можна побачити «світіння» цих структур (В). Анізотропні об’єкти роздвоюють промінь світла, що проходить через них, при цьому один промінь не змінює площину коливань і відсіюється аналізатором, інший змінює площину коливань і проходить через аналізатор, обумовлюючи «світіння» анізотропних структур на темному фоні. На даному препараті представлено кісткові балочки губчастої кістки тіла хребця. Забарвлення гематоксиліном та еозином. А – площини поляризації поляризатора і аналізатора співпадають; В – площини поляризації взаємно перпендикулярні. На зображенні В можна помітити «світіння» у периферійних ділянках кісткових балочок, обумовлене наявністю в органічному матриксі кістки колагену, якому притаманні анізотропні властивості. Між кістковими балочками розташовані клітини червоного кісткового мозку та кровоносні судини. Мікроскопія проведена за допомогою світлового мікроскопа Granum Trino R 6003 (DSM-310), об’єктив 10х, масштаб – 100 мкм. У якості поляризаційних фільтрів було використано дві поляризаційні плівки для екрану смартфонів, поляризатор було розташовані між конденсором та предметним столиком, аналізатор – між препаратом і об’єктивом.English: Photomicrograph of the vertebral body spongy bone, polarized light microscopy.
Polarized light microscopy differs from conventional light microscopy by the use of two polarizing filters – the polarizer and the analyzer, positioned between the light source and the specimen (polarizer) and between the specimen and the objective (analyzer). Both polarizing filters provide polarization of light in a specific plane, called the plane of polarization. When the planes of polarization of the polarizer and the analyzer coincide, polarized light that has passed through the polarizer goes through the specimen and freely passes through the analyzer (A). When the planes of polarization of the polarizer and the analyzer are mutually perpendicular, the light that has passed through the polarizer (polarized in the plane of the polarizer) is blocked by the analyzer, resulting in a dark background. However, if there are objects with anisotropic properties between the polarizer and the analyzer, the "illumination" of these structures can be observed (B). Anisotropic objects split the light beam passing through them, with one beam not changing the plane of oscillations and being blocked by the analyzer, and the other changing the plane of oscillations and passing through the analyzer, causing the "illumination" of anisotropic structures against a dark background. In this slide, bony trabeculae of the vertebral body spongy bone are presented. Hematoxylin and eosin staining. A – planes of polarization of the polarizer and the analyzer coincide; B – planes of polarization are mutually perpendicular. In image B, "illumination" can be noticed in the peripheral areas of the bone trabeculae, caused by the presence of collagen (exhibiting anisotropic properties) in the organic bone matrix. Red bone marrow cells and blood vessels are located between the bone trabeculae. Microscopy was conducted using the Granum Trino R 6003 (DSM-310) light microscope, 10x objective, scale bar – 100 µm. Two polarizing film sheets for smartphone screens were used as polarizing filters, the polarizer was positioned between the condenser and the stage, and the analyzer – between the specimen and the objective.English: Photomicrograph of the vertebral body spongy bone, polarized light microscopy. |
